Indications

Uniplar 1 is used for preventing and treating a condition called secondary hyperparathyroidism in people with Chronic Kidney Disease (CKD) in stages 3, 4, and 5. This includes adults and children aged 10 and older, especially for those on dialysis.

Pharmacology

Uniplar 1 is a synthetic version of vitamin D2 that helps reduce parathyroid hormone (PTH) levels. Uniplar 1 works by activating vitamin D pathways, which control the production of PTH, helping treat the condition linked with CKD.

Dosage & Administration

Adults & Elderly: Take one capsule daily or three times a week, depending on your doctor’s advice. If taking three times a week, space the doses every other day.

Starting Dose: The first dose depends on the patient’s PTH level. For lower levels of PTH (<500 pg/ml), the daily dose is 1 mcg, and three times a week Uniplar 1 is 2 mcg. For higher levels of PTH (>500 pg/ml), the daily dose is 2 mcg, and three times a week Uniplar 1 is 4 mcg.

Adjusting the Dose: Based on how the PTH level responds, the dose may be increased or decreased gradually. If the PTH level drops too much, the dose can be reduced.

Interaction

Taking Uniplar 1 with strong CYP3A inhibitors (like ketoconazole) can increase the drug’s effects. Avoid taking Uniplar 1 with cholestyramine or mineral oil as Uniplar 1 reduces absorption. Take Uniplar 1 at least 1 hour before or 4-6 hours after these medications.

Contraindications

Uniplar 1 should not be taken by people with high calcium levels or vitamin D toxicity.

Side effects may include high calcium levels, stomach problems, or other reactions. Consult a doctor if any unusual symptoms occur.

Pregnancy & Lactation

Pregnant women should consult their doctor before using Uniplar 1, as there may be risks. Breastfeeding is not recommended during treatment because Uniplar 1 might cause harm to the baby.

Precautions & Warnings

Watch out for high calcium levels, which can cause serious health issues. Use caution if taking other calcium-containing supplements or medications. Symptoms of high calcium include tiredness, nausea, and constipation.

Overdose Effects

Taking too much can cause high calcium levels, which may lead to serious problems like heart issues. If overdose happens, medical help should be sought immediately. Treatment may include stopping calcium supplements and adjusting the diet.
