Indications

Adinot is used to help people with alcohol addiction and to block the effects of opioids taken from outside the body. Adinot does not have any therapeutic benefit unless Adinot is part of a proper treatment plan for addiction.

Pharmacology

Adinot works by blocking opioid receptors in the body. This stops opioids from working and can cause withdrawal symptoms in people who are dependent on opioids.

Dosage & Administration

For opioid dependence: Start with 25 mg and increase to 50 mg per day if there are no withdrawal symptoms.

Maintenance: 350 mg weekly, either 50 mg daily or divided into 3 doses on different days of the week for better compliance.

For alcohol dependence: 50 mg per day.

Interaction

Adinot may reduce the effects of medications containing opiates, like those used for pain, cough, and diarrhea. Adinot can change the levels of other drugs that affect liver metabolism. Adinot may cause more damage to the liver when taken with disulfiram. Adinot can increase drowsiness and tiredness if used with thioridazine. Adinot may also change insulin requirements.

Contraindications

Adinot should not be used by people who are also dependent on opioids, have acute hepatitis or liver failure, or are experiencing acute opioid withdrawal. Adinot should not be used by patients taking opioid painkillers.

Side Effects

Possible side effects include stomach pain, nausea, vomiting, anxiety, trouble sleeping, tiredness, headache, muscle pain, loss of appetite, diarrhea, constipation, increased thirst, chest pain, chills, dizziness, sexual problems, skin rash, liver issues, and low platelet count. Reactions at injection sites may also occur.

Pregnancy & Lactation

Adinot is classified as Pregnancy Category C. Animal studies have shown negative effects on the fetus, and there are no controlled studies in pregnant women. Adinot should only be used if the potential benefits outweigh the risks to the baby.

Precautions & Warnings

Patients with liver or kidney problems should be careful when using this medicine. Liver function tests should be monitored regularly. Patients should be free from opioids for at least 7-10 days before starting naltrexone. They should also be warned against using opioids while on naltrexone. Monitoring for injection site reactions is advised. Adinot should be used cautiously during pregnancy, breastfeeding, and in those with a history of bleeding disorders or low platelet count.

Overdose Effects

Symptoms of overdose include seizures and trouble breathing.

Management: Supportive care and symptomatic treatment should be provided.
