Indications
Ajuben 12 is used to treat chorea related to Huntington’s disease and tardive dyskinesia in adults.
Pharmacology
Ajuben 12 works by reducing levels of certain chemicals (like dopamine, serotonin, norepinephrine, and histamine) in the brain. This happens because Ajuben 12 helps deplete these chemicals from nerve endings. The main substances formed from Ajuben 12 in the body also reduce the uptake of these chemicals, leading to lower levels in the brain.
Dosage & Administration
For patients not taking Tetrabenazine:
Chorea with Huntington’s disease:
Start with 6 mg per day. The dose can go up to 48 mg per day in divided doses.
Tardive dyskinesia:
Start with 12 mg per day. The dose can go up to 48 mg per day in divided doses.
Adjust doses weekly by 6 mg until a dose that works is found, but do not exceed 48 mg per day.
For patients switching from Tetrabenazine:
Start the next day with the following initial dose of deutetrabenazine based on the current dose of tetrabenazine:
- 12.5 mg tetrabenazine → 6 mg once daily
- 25 mg tetrabenazine → 6 mg once daily
- 37.5 mg tetrabenazine → 9 mg once daily
- 50 mg tetrabenazine → 12 mg once daily
- 62.5 mg tetrabenazine → 15 mg once daily
- 75 mg tetrabenazine → 18 mg once daily
- 87.5 mg tetrabenazine → 21 mg once daily
- 100 mg tetrabenazine → 24 mg once daily
Adjust the dose weekly after switching to deutetrabenazine.
Interaction
Ajuben 12 should not be taken with strong CYP2D6 inhibitors. The maximum recommended dose in these cases is 36 mg per day. Be cautious when using alcohol or sedating drugs, as they may increase drowsiness and sedation.
Contraindications
Ajuben 12 is not suitable for people with suicidal tendencies, untreated or inadequately treated depression, liver issues, or those taking MAOIs, reserpine, or tetrabenazine.
Side Effects
Common side effects include drowsiness, diarrhea, dry mouth, and tiredness.

Precautions & Warnings

Overdose Effects
Overdosing may cause symptoms like muscle spasms, nausea, vomiting, sweating, low blood pressure, confusion, diarrhea, hallucinations, and shaking.
