Indications
Ambeexin is used to treat infections caused by bacteria that are sensitive to ß-lactamase negative strains. These infections include ear, nose, and throat infections like otitis media, sinusitis, tonsillitis, pharyngitis, and laryngitis. Ambeexin also treats lower respiratory tract infections such as pneumonia, bronchitis, lung abscess, empyema, and bronchiectasis. Ambeexin is used for skin and soft tissue infections like cellulitis, carbuncles, and infected wounds. Ambeexin can treat genito-urinary tract infections such as pyelonephritis, cystitis, and urethritis, as well as venereal diseases like acute uncomplicated gonorrhoea. For dental abscess, Ambeexin is used for short-term therapy. Additionally, Ambeexin is used with Clarithromycin and Lansoprazole in triple therapy for treating H. pylori infection and duodenal ulcer disease, and to reduce the risk of duodenal ulcer recurrence.
Pharmacology
Ambeexin is a broad-spectrum penicillin effective against a wide range of Gram-positive and Gram-negative bacteria. Ambeexin works by inhibiting the production of the bacterial cell wall. Ambeexin remains stable in stomach acid and is absorbed quickly after taking Ambeexin orally. After taking an oral dose, peak blood levels are reached within 1 to 2 hours. Ambeexin is well-distributed in various body tissues and fluids.
Dosage & Administration
Adults: 250 mg three times a day, increasing to 500 mg three times a day for more severe infections.
Children (up to 10 years old): 125 mg three times a day, increasing to 250 mg three times a day for severe infections. For severe or recurrent respiratory infections, 3 gm every 12 hours is recommended.
Otitis Media: For adults, 1 g three times a day; for children, 40 mg/kg body weight daily in 3 divided doses (max. 3 g daily).
Pneumonia: 500-1000 mg three times a day is recommended.
Dental Abscess: 3 gm, repeated after 10-12 hours.
Urinary Tract Infections: 3 gm, repeated after 10-12 hours.
Gonorrhoea: A single dose of 2-3 gm with 1 gm Probenecid is recommended (Probenecid is not for children under 2 years old).
In Renal Impairment: Dosage may need to be reduced.
Interaction
Taking Ambeexin with Probenecid may increase and prolong blood levels. Ambeexin can affect the gut bacteria and reduce the effectiveness of combined oral contraceptives that contain estrogen and progesterone.
Contraindications
Ambeexin should not be used in patients who are allergic to penicillin.
Side Effects
Side effects are generally mild and temporary. These may include diarrhea, indigestion, or occasionally a rash. Rarely, pseudo-membranous colitis has been reported.
Pregnancy & Lactation
Ambeexin is classified as pregnancy category B by the FDA. There are no adequate and well-controlled studies in pregnant women, so Ambeexin should be used during pregnancy only if clearly needed. Ambeexin is excreted in human milk, so caution is advised when used by lactating mothers.
Precautions & Warnings
Superinfections with fungal or bacterial pathogens may occur during therapy. If this happens, Ambeexin should be discontinued, and the appropriate treatment should be started.
