Indications
Corestin 5 is used to help lower bad cholesterol (LDL-C) and increase good cholesterol (HDL-C) in patients with high cholesterol levels. Corestin 5 is also used to help lower triglyceride levels and can be given to those with certain types of high blood fat levels. Corestin 5 is also used for patients with primary dysbetalipoproteinemia, as an addition to diet.
Pharmacology
Corestin 5 works by blocking a substance in the liver that is needed to make cholesterol. This helps to lower cholesterol levels in the blood. Corestin 5 also helps to increase the number of receptors in the liver to remove bad cholesterol from the blood. This reduces the amount of bad cholesterol and fat in the bloodstream.
Dosage & Administration
For children with Familial Hypercholesterolemia (ages 10-17): The usual dose is 5-20 mg/day, with a maximum dose of 20 mg/day. Doses should be adjusted based on the target cholesterol levels.
For patients with very high cholesterol or a high risk of heart disease or stroke: 40 mg once daily.
For patients with Homozygous Familial Hypercholesterolemia: The starting dose is 20 mg once daily. The response should be checked using cholesterol levels before treatments like apheresis.
For Asian patients: The starting dose should be 5 mg once daily.
In patients taking Cyclosporine, Lopinavir/Ritonavir, or Atazanavir/Ritonavir: The dose should be limited to 5 mg once daily for Cyclosporine and 10 mg once daily for Lopinavir/Ritonavir or Atazanavir/Ritonavir.
In patients with severe kidney issues: The starting dose should be 5 mg once daily, not exceeding 10 mg daily.
Interaction
Corestin 5 may interact with certain drugs such as Cyclosporine, Gemfibrozil, and Lopinavir/Ritonavir, increasing the amount of the medicine in the bloodstream. Combining Corestin 5 with other cholesterol-lowering drugs, like Niacin and Fibrates, may increase the risk of muscle problems. Corestin 5 may also affect the blood’s clotting time if taken with blood thinners like Coumarin anticoagulants.
Contraindications
Corestin 5 should not be used by individuals who are allergic to any of its components, have liver diseases, or have unexplained liver problems. Corestin 5 is not recommended for pregnant women or those who are breastfeeding.
Side Effects
Common side effects may include headache, muscle pain, constipation, tiredness, stomach pain, and nausea. These effects are usually mild and go away on their own.
Pregnancy & Lactation
Corestin 5 should only be used in women who are unlikely to get pregnant. If a woman becomes pregnant while taking Corestin 5, she should stop the treatment immediately. Corestin 5 is unknown whether Corestin 5 passes into breast milk, so caution should be taken if breastfeeding.
Precautions & Warnings
Corestin 5 should be used carefully in people who are at higher risk for muscle problems, including older adults, those with untreated thyroid issues, or kidney problems. If muscle pain or weakness occurs, Corestin 5 may be a sign of a serious condition and should be reported to a doctor immediately. Regular liver tests are recommended during treatment to monitor for any potential issues.
Overdose Effects
There is no specific treatment for overdose. If an overdose occurs, the patient should be treated to manage symptoms, and supportive care should be provided. Hemodialysis does not significantly help clear the medicine from the body.
