Indications

Crizonix is used for treating metastatic non-small cell lung cancer (NSCLC) in patients whose tumors are positive for anaplastic lymphoma kinase (ALK) or ROS1.

Pharmacology

Crizonix works by blocking certain proteins (like ALK, c-Met, and ROS1) that help tumors grow. By inhibiting these proteins, Crizonix helps stop cancer cells from spreading and can reduce tumor size. Crizonix has shown effectiveness in reducing tumors in laboratory studies and animal models.

Dosage & Administration

Recommended Dose: 250 mg taken orally, twice a day.

Geriatric Use: No difference in safety or effectiveness was found between younger and older patients.

Pediatric Dose: Its safety and effectiveness for children have not been established.

Renal Impairment: In patients with severe kidney problems, take 250 mg once a day. No adjustment needed for mild or moderate kidney problems.

Hepatic Impairment: Use with caution in patients with liver issues.

Interaction

Crizonix should not be used with strong CYP3A inhibitors like atazanavir or ketoconazole. Avoid using Crizonix with CYP3A inducers like carbamazepine or St. John’s Wort. Crizonix should also be avoided with other CYP3A substrates that have narrow therapeutic ranges, such as cyclosporine or fentanyl.

Side Effects

Common side effects include vision problems, nausea, diarrhea, vomiting, swelling, constipation, tiredness, decreased appetite, dizziness, and neuropathy. Elevated liver enzymes may also occur.

Pregnancy & Lactation

Crizonix can harm the fetus if taken during pregnancy. Women who are pregnant or planning to become pregnant should avoid Crizonix. Crizonix is not known whether Crizonix passes into breast milk, so breastfeeding is not recommended during treatment and for 45 days after the last dose.

Precautions & Warnings

Patients should have regular liver tests. Crizonix may need to be paused or stopped if there is liver damage. Crizonix may cause lung issues, so if this happens, Crizonix should be permanently stopped. People with a history of heart problems should be monitored for slow heart rate. Crizonix can cause severe vision loss, and treatment should be stopped if this occurs.
