Indications
Cykin 125 is used to treat advanced or metastatic breast cancer that is hormone receptor-positive and HER2-negative. Cykin 125 is combined with an aromatase inhibitor for postmenopausal women or Fulvestrant in women who have had disease progression after endocrine therapy.
Pharmacology
Cykin 125 works by inhibiting Cyclin-dependent kinases (CDK) 4 and 6, which are involved in cell growth. Cykin 125 blocks the progression of cancer cells in estrogen receptor-positive breast cancer by stopping the cell cycle at an early stage. When combined with antiestrogen treatments, Cykin 125 increases the growth arrest and helps control tumor growth more effectively than when used alone.
Dosage & Administration
Adult & Elderly: Take 125 mg capsule once daily for 21 days, followed by 7 days off treatment. This completes a 28-day treatment cycle. Cykin 125 should be taken with food at the same time each day.
Recommended Dose Modifications: If side effects occur, the dose can be reduced to 100 mg/day or 75 mg/day. If further reduction is needed, discontinue use, or follow the physician’s instructions.
Use in Children: Cykin 125 is not recommended for use in children.
Interaction
Cykin 125 is processed by the liver enzymes CYP3A and Sulfotransferase. Avoid taking Cykin 125 with strong CYP3A inhibitors like Itraconazole, as this can increase its levels in the body. Cykin 125 is also advised not to consume grapefruit juice during treatment. On the other hand, certain drugs like Rifampin can lower its effectiveness, so avoid combining Cykin 125 with strong CYP3A inducers.

Side Effects
Common side effects include lowered white blood cell counts, neutropenia, infections, fatigue, nausea, hair loss, mouth inflammation, diarrhea, anemia, rash, and fever.
Pregnancy & Lactation
If pregnant or breastfeeding, consult a doctor before taking Cykin 125. Cykin 125 can harm a fetus, and there is a risk of birth defects and miscarriage in pregnant women.
Precautions & Warnings
Patients should be monitored for low white blood cell counts (neutropenia). Severe cases of lung disease (ILD) and breathing problems (pneumonitis) have been reported, so patients should be checked for symptoms such as cough or shortness of breath. Cykin 125 can also harm an unborn child, so effective contraception is advised during treatment and for at least 3 weeks after stopping Cykin 125.
Overdose Effects
There is no specific antidote for overdose. Supportive care is the recommended treatment in case of overdose.
