Indications

D-Zole 1gm is used for treating intestinal amoebiasis, hepatic amoebiasis, urethritis and vaginitis caused by Trichomonas vaginalis, and giardiasis.

Pharmacology

D-Zole 1gm is quickly absorbed when taken by mouth, with the highest blood level reached 3 hours after taking a 2 gm dose. The body eliminates D-Zole 1gm mainly through urine, with about half of the dose leaving the body in 5 days. D-Zole 1gm has the longest half-life of second-generation nitroimidazoles, which allows D-Zole 1gm to maintain therapeutic blood levels for up to 72 hours after a single dose.

Dosage & Administration

Acute Intestinal Amoebiasis:
Adults: 2 gm as a single dose, preferably before a meal.
Children: 30 mg/kg as a single dose, preferably before a meal.

Asymptomatic Amoebiasis (minute and cystic form):
Adults: 2 gm once a day for 3 days, preferably before a meal.
Children: 30 mg/kg once a day for 3 days, preferably before a meal.

Hepatic Amoebiasis:
Pus removal should be done along with treatment.
Adults: 1.50 gm/day in single or divided doses, before meals, for 5 days.
Children: 30 mg/kg/day, in single or divided doses, before meals, for 5 days.

Giardiasis:
Adults: 2 gm as a single dose, preferably before a meal.
Children: 35-50 mg/kg as a single dose, preferably before a meal.

Trichomoniasis:
Adults: 2 gm as a single dose, preferably before a meal. The partner should also take the same treatment at the same time.

Interaction

D-Zole 1gm should not be taken with disulfiram, as this may cause confusion and paranoia. When used with warfarin, monitoring is necessary because D-Zole 1gm may increase the effects of anticoagulants and raise the risk of bleeding.

Contraindications

D-Zole 1gm is not recommended for patients who are allergic to imidazole derivatives.

Side Effects

D-Zole 1gm is generally well tolerated, with few serious side effects. However, some people may experience:

Common side effects: Stomach issues like nausea, pain, metallic taste, and mouth irritation.

Occasional side effects: Skin reactions like hives, and mild decrease in white blood cells that goes away once the treatment stops.

Rare side effects: Dizziness, coordination problems, numbness, or nerve damage.

Pregnancy & Lactation

D-Zole 1gm may be used during pregnancy after the first trimester. D-Zole 1gm should not be used during the first trimester or while breastfeeding, as D-Zole 1gm passes into the placenta and breast milk.

Precautions & Warnings

D-Zole 1gm is important not to drink alcohol while using this medicine due to the risk of a severe reaction. D-Zole 1gm should also be avoided by patients with a history of blood disorders.
