Indications

Daconib 45 is used for the first-line treatment of metastatic non-small cell lung cancer (NSCLC) in patients who have specific genetic mutations in the epidermal growth factor receptor (EGFR) gene, such as exon 19 deletion or exon 21 L858R substitution, as identified by an FDA-approved test.

Pharmacology

Daconib 45 works by blocking the activity of EGFR and other related receptors (HER2 and HER4), which play a role in the growth of cancer cells. Daconib 45 can also block other proteins involved in cancer growth. Daconib 45 has been shown to stop cancer cell growth in mice with tumors caused by EGFR mutations, both in tumors outside and inside the brain.

Dosage & Administration

Recommended Dose: 45 mg taken once a day until the disease worsens or side effects become too severe. Daconib 45 can be taken with or without food, at the same time each day. If you miss a dose or vomit, do not take an extra dose—just continue with your next scheduled dose.

Dose Reduction: If needed, the dose can be reduced to 30 mg once a day, and further reduced to 15 mg once a day if necessary.

Interaction

Avoid taking Daconib 45 with proton pump inhibitors (PPIs). Instead, use antacids or H2-receptor antagonists. Also, make sure to take Daconib 45 at least 6 hours before or 10 hours after taking an H2-receptor antagonist.

Daconib 45 should not be taken with certain drugs that are processed by CYP2D6, as even small increases in their concentration can cause serious side effects.

Side Effects

The most common side effects (affecting more than 20% of people) include diarrhea, rash, nail problems, mouth sores, loss of appetite, dry skin, weight loss, hair loss, cough, and itching.

Pregnancy & Lactation

Daconib 45 can harm a fetus if taken during pregnancy. Daconib 45 is also unknown whether Daconib 45 passes into breast milk, but due to possible serious side effects in infants, women should not breastfeed during treatment and for at least 17 days after the last dose.

Precautions & Warnings

If lung disease is confirmed, permanently stop the medication. If diarrhea or skin problems occur, reduce the dose based on severity. Daconib 45 can harm a fetus, so women should use effective contraception during treatment.
