Indications
Danafil is used to treat men with erectile dysfunction, which is the inability to achieve or maintain an erection for satisfactory sexual performance. For Danafil to work, sexual stimulation is needed. Danafil is also used to treat pulmonary arterial hypertension.
Pharmacology
Danafil works by increasing the levels of cGMP in the penis when sexual stimulation occurs. This helps relax the muscles and allows more blood to flow into the penis. Danafil does this by blocking the enzyme PDE5, which normally breaks down cGMP. Danafil has no effect on its own without sexual stimulation. The body processes Danafil mainly in the liver, and Danafil lasts about 4 hours in the system.
Dosage & Administration
Erectile Dysfunction: The usual dose is 50 mg, taken about 1 hour before sexual activity. Danafil can be taken anytime between 4 hours and 30 minutes before sex. The dose can be adjusted between 25 mg and 100 mg depending on effectiveness and tolerance. Danafil can be taken once a day at most.
Pulmonary Arterial Hypertension: The usual dose is 20 mg, taken three times a day, with or without food.
Interaction
Danafil is mainly processed by certain enzymes in the liver. Drugs that affect these enzymes can change how Danafil works. Some medicines, like ketoconazole and erythromycin, can increase its levels in the body, while others, like rifampin, can decrease its levels. Danafil should not be taken with nitrates or nitric oxide donors, as this can lower blood pressure too much. Taking Danafil with some other medications, like alpha-blockers, may also lower blood pressure and cause dizziness.
Contraindications
Danafil should not be used by anyone allergic to any of its ingredients or by those taking nitrates, as Danafil can dangerously lower blood pressure.
Side Effects
Common side effects include face swelling, headache, dizziness, and stomach pain. Danafil can also cause heart problems, vision issues, and prolonged or painful erections. Serious effects like heart attack, stroke, or seizures can occur, though these are rare. Consult a doctor if any of these happen.
Pregnancy & Lactation
Danafil is not intended for use in women and should not be used during pregnancy or breastfeeding. Animal studies have not shown harm, but there is no clear evidence in humans.
