Indications

Denosis 60 is used to treat postmenopausal women with osteoporosis who are at high risk for fractures. Denosis 60 helps reduce the chance of fractures in the spine, hips, and other bones. Denosis 60 is also used to help men with osteoporosis, and people whose bone loss is caused by taking steroids or hormone treatments for prostate or breast cancer. Additionally, Denosis 60 is used to prevent bone problems in people with multiple myeloma or bone metastasis, and to treat giant cell tumors of bone.

Pharmacology

Denosis 60 is a monoclonal antibody that targets a protein called RANKL. RANKL is important for bone resorption, which is the process of breaking down bone tissue. By blocking RANKL, Denosis 60 reduces the activity of cells that break down bones, helping to build and maintain stronger bones.

Dosage & Administration

Osteoporosis and Bone Loss: 60 mg injected every 6 months into the upper arm, thigh, or abdomen. Patients should also take 1000 mg of calcium and 400 IU of vitamin D daily. If a dose is missed, Denosis 60 should be taken as soon as possible, then scheduled for every 6 months.

Multiple Myeloma and Bone Metastasis: 120 mg injected every 4 weeks.

Giant Cell Tumor of Bone: 120 mg injected every 4 weeks with extra doses on days 8 and 15 during the first month of treatment.

Hypercalcemia of Malignancy: 120 mg injected every 4 weeks, with extra doses on days 8 and 15 of the first month of treatment.

Interaction

Denosis 60 does not affect the metabolism of midazolam, which is processed by a liver enzyme (CYP3A4). This suggests Denosis 60 does not interact with drugs that are metabolized by this enzyme in patients with postmenopausal osteoporosis.

Contraindications

Denosis 60 should not be used in people who have low calcium levels (hypocalcemia) until their calcium levels are corrected. Denosis 60 should also not be used in pregnant women, as Denosis 60 may harm the fetus, and in people with a history of severe allergic reactions to any of its ingredients.

Side Effects

Common side effects include back pain, pain in limbs, musculoskeletal pain, and high cholesterol. Some people may experience more serious effects like infections, bone pain, or low blood calcium levels. Severe reactions include allergic reactions, jaw problems, and fractures.

Pregnancy & Lactation

Denosis 60 may harm an unborn baby, so Denosis 60 should not be used during pregnancy. Women who are planning to become pregnant should discuss their treatment with a doctor. If breastfeeding, consider the benefits and risks of using Denosis 60 while nursing.

Precautions & Warnings

People with low calcium levels should correct them before starting treatment. Denosis 60 can cause serious side effects like jaw problems (osteonecrosis) and unusual bone fractures. Patients should have regular check-ups and inform their doctor about any signs of infection or bone pain. Denosis 60 is not recommended to stop the treatment suddenly without switching to another bone-strengthening medicine.

Storage Conditions
