Indications

Don A is used to treat symptoms like bloating, stomach discomfort, and indigestion. Don A helps with nausea, vomiting, and heartburn. Don A is also used for Parkinson’s disease-related nausea and to speed up barium movement in certain medical scans.

Pharmacology

Don A is a medication that works by blocking certain receptors in the stomach and brain. This helps improve stomach movement, speeds up digestion, and reduces nausea. Don A mostly works in the stomach, with little effect on the brain, so Don A doesn’t cause mental side effects. Don A also helps prevent stomach contents from coming back up.

Dosage & Administration

Adults: Take 10-20 mg (1-2 tablets or 10-20 ml suspension) every 6-8 hours. The maximum daily dose is 80 mg.

Children: Take 2-4 ml suspension per 10 kg of body weight every 6-8 hours.

For acute conditions: Adults take 20 mg (2 tablets or 20 ml suspension) every 6-8 hours. Children take 0.2-0.4 mg/kg of body weight every 6-8 hours.

By rectum (suppositories): Adults take 30-60 mg every 4-8 hours. Children (10-25 kg) take a maximum of 30 mg per day, divided if necessary. Treatment should not last longer than 12 weeks.

Interaction

Don A should not be taken with certain antacids or anticholinergic drugs, as they reduce its effect. Don A is processed in the body by a specific enzyme, so taking other medications that affect this enzyme may increase its level. Don A can also influence the absorption of other oral medications, especially those with slow-release or special coatings.

Contraindications

Don A should not be used by people who are allergic to Don A or by newborns. Don A should not be used in certain gastrointestinal problems, such as bleeding or blockages. Don A is also not recommended for people with certain types of tumors in the pituitary gland.

Side Effects

Side effects are rare, but may include temporary stomach cramps, dizziness, or skin reactions like rashes. In very rare cases, Don A may cause an increase in a hormone that can lead to milk production in both men and women, or swelling in men’s breasts. If used in infants or people with brain-related conditions, there could be a risk of neurological side effects.

Pregnancy & Lactation

Don A has not caused birth defects in animals, but Don A should only be used during pregnancy if absolutely necessary. Don A passes into breast milk, so Don A is not recommended for breastfeeding mothers unless the benefits outweigh the risks.

Precautions & Warnings

Don A should be used cautiously in children and in people with liver problems. Young children may have a higher risk of certain side effects because their brain barrier isn’t fully developed.

Overdose Effects

Overdose symptoms may include drowsiness, confusion, or movement problems, especially in children. If an overdose occurs, activated charcoal and monitoring of the patient are recommended. Medications for movement problems may also be used if necessary.

Therapeutic Class

Don A is a motility stimulant and a dopamine antagonist, used to help with stomach movement and related issues.

Storage Conditions

Store below 30°C, away from light and moisture. Keep out of reach of children.
