Indications
Loperin is indicated for the treatment of acute and chronic diarrhea, abdominal cramps associated with diarrhea, Crohn’s disease, post-gastric surgery, and ulcerative colitis.
Pharmacology
Loperin works by reducing the movement of muscles in the intestine, which helps slow down diarrhea. Loperin does this by binding to opiate receptors in the gut, increasing the time Loperin takes for the intestines to move food along. Loperin also increases the tone of the anal sphincter and reduces fluid and electrolyte loss in the intestines.
After being taken, Loperin is absorbed in the body at a rate of more than 65%. Peak levels are reached about 4 hours after taking the medication. Loperin undergoes metabolism in the liver and intestines, with only a small amount entering the bloodstream. Loperin is eliminated mostly through the bile and feces.
Dosage & Administration
Acute diarrhea:
Adults: 2 capsules initially, then 1 capsule after each loose stool. The usual dose is 3-4 capsules a day. The maximum daily dose should not exceed 8 capsules.
Children (9-12 years): 1 capsule after each loose stool until diarrhea stops, but not more than 6 capsules daily.
Children (5-9 years): 1 capsule 4 times a day until diarrhea stops, but not more than 4 capsules daily.
Chronic diarrhea:
Adults: 2-4 capsules daily, divided into doses based on severity.
Children: 1-2 capsules daily based on severity, or as directed by a doctor.

Contraindications
Loperin is contraindicated in patients with hypersensitivity to the drug or those who must avoid constipation. Loperin should not be used alone in cases of acute dysentery, where there is blood in the stool and elevated body temperature.
Side Effects
Side effects may include abdominal pain, dry mouth, dizziness, fatigue, skin rashes, and other gastrointestinal issues.
Pregnancy & Lactation
Loperin is not recommended during pregnancy unless necessary. Loperin has been shown that only a small amount passes into breast milk, but caution is advised if used by nursing mothers.
Precautions & Warnings
Loperin should be used with caution in patients with severe liver problems.
Overdose Effects
Overdosing may cause constipation, CNS depression, and nausea.
