Indications

Ranidin is used for the treatment of:

  • Active duodenal ulcer
  • Benign gastric ulcer
  • Treatment and prevention of ulcers caused by non-steroidal anti-inflammatory drugs (NSAIDs)
  • Post-operative stress ulcers
  • Zollinger-Ellison Syndrome
  • Gastroesophageal reflux disease (GERD)
  • Gastrointestinal bleeding from stress ulcers in seriously ill patients
  • Recurrent bleeding in patients with bleeding peptic ulcers
  • Before general anesthesia in patients at risk of acid aspiration, especially pregnant women

Pharmacology

Ranidin works by blocking histamine at the H2-receptors in the stomach, reducing the production of gastric acid. Ranidin does not affect the production of pepsin, intrinsic factors, or serum gastrin.

Dosage & Administration

For Tablets & Syrup:

  • Duodenal and gastric ulcers: 150 mg twice daily, or 300 mg once at night, for 4 to 8 weeks.
  • Reflux esophagitis: 150 mg twice daily, or 300 mg at bedtime, for up to 8 weeks.
  • Zollinger-Ellison syndrome: 150 mg three times daily, increased to up to 6 g daily in divided doses.
  • Episodic dyspepsia: 150 mg twice daily, or 300 mg at bedtime, for up to 6 weeks.
  • Maintenance: 150 mg at night to prevent recurrence.
  • Children (peptic ulcer): 2-4 mg/kg twice daily, with a maximum of 300 mg daily.

For IV Injection & Infusion:

  • 50 mg intravenous injection, diluted to 20 ml, every 6 to 8 hours or a 25 mg/hour infusion for two hours, repeated every 6 to 8 hours.
  • For prevention of gastrointestinal bleeding in seriously ill patients, 50 mg intravenous injection followed by a continuous infusion of 0.125-0.250 mg/kg/hour.
  • If at risk for aspiration syndrome, administer 50 mg intramuscularly or by slow intravenous injection 45 to 60 minutes before anesthesia.

Children: The recommended oral dose for treating peptic ulcer is 2 mg/kg to 4 mg/kg twice daily, up to a maximum of 300 mg per day. Safety of the IV injection in children has not been established.

Interaction

Taking Ranidin with propantheline bromide may delay absorption and increase peak serum levels. Ranidin has a minor effect on the liver metabolism of certain drugs like anticoagulants, theophylline, diazepam, and propranolol. Ranidin may also affect the absorption of drugs dependent on stomach pH, such as ketoconazole and glipizide, and reduce the bioavailability of antacids.

Contraindications

Ranidin should not be used in patients who are allergic to Ranidin.

Side Effects

Side effects are uncommon, but some people may experience mild issues such as dizziness, rash, tiredness, confusion, headache, muscle or joint pain, and changes in bowel movements. These side effects are usually temporary.

Pregnancy & Lactation

Pregnancy: Ranidin can cross the placenta, but there is no evidence that Ranidin harms the baby. Ranidin should only be used during pregnancy if absolutely necessary.

Lactation: Ranidin is passed into breast milk. Caution is advised if taken by a nursing mother.

Precautions & Warnings

Patients with kidney or liver problems may require a lower dose. Always consult your doctor before using Ranidin if you have these conditions.

Overdose Effects

Overdosing is usually not a major concern, but if Ranidin happens, supportive treatment should be given. If necessary, the drug can be removed from the bloodstream using dialysis.

Ranidin belongs to the class of H2 receptor antagonists.

Store in a cool, dry place away from light. Keep Ranidin out of the reach of children.
