Indications
Safyron 10 is used to treat conditions where there is a lack of progesterone, such as:
- Threatened miscarriage
- Habitual miscarriage
- Dysmenorrhoea (painful periods)
- Endometriosis
- Secondary amenorrhoea (lack of periods)
- Irregular menstrual cycles
- Dysfunctional uterine bleeding
- Infertility due to luteal insufficiency
- Luteal support in Assisted Reproductive Technology (ART)
Safyron 10 is also used in hormone replacement therapy to protect the uterus from the effects of estrogen after menopause, whether caused naturally or surgically.
Pharmacology
Safyron 10 is an oral medication that acts as a progestogen. Safyron 10 helps make the lining of the uterus ready to receive a fertilized egg. Safyron 10 provides protection against the risk of endometrial problems (such as cancer) that can be caused by estrogen. Safyron 10 is used when there is a deficiency of natural progesterone. Safyron 10 has no estrogen, androgen, or other hormonal effects.
Dosage & Administration
Always take the 10 mg tablet as prescribed by your doctor.
For progesterone deficiencies:
- Threatened miscarriage: 40 mg (four tablets) at once, then 10 mg (one tablet) every eight hours until symptoms go away.
- Habitual miscarriage: 10 mg (one tablet) twice a day until the twentieth week of pregnancy.
- Dysmenorrhoea: 10 mg (one tablet) twice a day from day 5 to day 25 of the menstrual cycle.
- Endometriosis: 10 mg (one tablet) two or three times a day from day 5 to day 25 of the cycle or continuously.
- Dysfunctional bleeding (to stop bleeding): 10 mg (one tablet) twice a day for five to seven days, combined with estrogen.
- Dysfunctional bleeding (to prevent bleeding): 10 mg (one tablet) twice a day from day 11 to day 25 of the cycle, combined with estrogen.
- Amenorrhoea: Take an estrogen once daily from day 1 to day 25 of the cycle, with 10 mg of this medication twice daily from day 11 to day 25.
- Irregular cycles: 10 mg (one tablet) twice a day from day 11 to day 25 of the cycle.
- Infertility due to luteal insufficiency: 10 mg (one tablet) daily from day 14 to day 25 of the cycle. Continue for at least 6 months. Continue this treatment during the first month of pregnancy if necessary.
- Luteal support in ART: 10 mg three times a day, starting from the day of egg retrieval and continuing for 10 weeks if pregnancy occurs.
- Hormone Replacement Therapy: Take one tablet daily for 14 days as part of a 28-day cycle, combined with estrogen. Consult your doctor if unsure about your estrogen therapy.

Contraindications
Safyron 10 should not be used by individuals who are allergic to this medication or any of its ingredients. Safyron 10 is also contraindicated in those with progestogen-dependent neoplasms (such as meningioma).
Side Effects
The most common side effects are headaches, nausea, menstrual issues, and breast pain or tenderness.
Pregnancy & Lactation
More than 10 million pregnancies have been exposed to this medication, and no harmful effects have been reported. There is no information on whether Safyron 10 passes into breast milk, but similar medications have been shown to do so in small amounts. Therefore, Safyron 10 should not be used while breastfeeding.
Precautions & Warnings
Before starting treatment, the cause of abnormal bleeding should be determined. Spotting or breakthrough bleeding may occur during the first months of use. If this continues, further investigation is needed. Treatment should be stopped if any of the following occur:
- Severe headache or migraine
- High blood pressure
- Venous thromboembolism (VTE)
Special monitoring is required during treatment for women with a history of depression.
Overdose Effects
Overdose is usually well-tolerated, with a maximum daily dose of 360 mg. If an overdose is suspected within 2-3 hours, gastric lavage is recommended. There are no specific antidotes, and treatment should be symptomatic.
