Indications
Tasso 80 is used for the first-line treatment of metastatic non-small cell lung cancer (NSCLC) in patients whose tumors have specific mutations in the epidermal growth factor receptor (EGFR), such as exon 19 deletions or exon 21 L858R mutations. Tasso 80 is also used to treat patients with metastatic EGFR T790M mutation-positive NSCLC whose disease has worsened after EGFR tyrosine kinase inhibitor (TKI) therapy.
Pharmacology
Tasso 80 is a kinase inhibitor that targets the epidermal growth factor receptor (EGFR). Tasso 80 irreversibly binds to mutant forms of EGFR, including T790M, L858R, and exon 19 deletions. Tasso 80 has shown anti-tumor effects in laboratory and animal models against NSCLC with EGFR mutations. Two active metabolites, AZ7550 and AZ5104, also contribute to its effects, with AZ5104 being more potent against certain EGFR mutations. The drug is absorbed in the body, and its levels increase proportionally with the dose. After taking Tasso 80 daily, steady levels are reached in about 15 days.
Dosage & Administration
The recommended dose is 80 mg once a day. Tasso 80 can be taken with or without food. If you miss a dose, do not take an extra dose to make up for Tasso 80. For patients who need to take the tablet with water, dissolve Tasso 80 in 60 ml of non-carbonated water, stir until Tasso 80 is in small pieces (not completely dissolved), and swallow Tasso 80 immediately. Do not crush, heat, or ultrasonicate the tablet. If using a nasogastric tube, dissolve the tablet in 15 ml of non-carbonated water and transfer the mixture to the syringe. Use an additional 15 ml of water to rinse and administer.
Interaction
Avoid using Tasso 80 with strong CYP3A inhibitors like certain antibiotics (e.g., Telithromycin), antifungals (e.g., Itraconazole), and antivirals (e.g., Ritonavir) as they may increase its concentration in the body. Also, avoid using Tasso 80 with CYP3A inducers (e.g., Phenytoin, Rifampicin) as they may decrease its effectiveness. Be cautious when taking Tasso 80 with drugs that interact with CYP3A, BCRP, or CYP1A2.

Side Effects
Common side effects (in at least 20% of patients) include diarrhea, skin rash, dry skin, nail problems, mouth sores, fatigue, and loss of appetite.
Pregnancy & Lactation
Tasso 80 can harm the fetus, so Tasso 80 should not be used during pregnancy unless absolutely necessary. There is no information about whether Tasso 80 passes into breast milk, so if you are breastfeeding or planning to breastfeed, consult your doctor. Women should use effective contraception during treatment and for 6 weeks afterward. Men should use contraception during treatment and for 4 months after finishing the drug.
Precautions & Warnings
There is a risk of interstitial lung disease (ILD)/pneumonitis in a small percentage of patients. Tasso 80 may also cause QTc interval prolongation, so monitor heart activity in patients with a history of heart problems. Cardiomyopathy has been observed in some patients, and Tasso 80 is important to monitor heart function regularly. Tasso 80 can also harm the fetus, so women should be cautious and use contraception during and after treatment.
