Indications
Vaxitet Ampoule is used to prevent tetanus in children 7 years of age or older and adults, especially those at risk of tetanus infection, like gardeners, farm workers, and athletes. Vaxitet Ampoule is also used to prevent neonatal tetanus by vaccinating women of childbearing age and to prevent tetanus after injury. Vaxitet Ampoule can be safely given at the same time as other vaccines such as BCG, Measles, Polio, Hepatitis B, Yellow fever, Haemophilus influenzae-B, and Varicella vaccines.
For people with HIV: Vaxitet Ampoule may be used in children with known or suspected HIV infection, though studies are still ongoing. There is no evidence yet of any increased side effects in children with HIV.
Pharmacology
Vaxitet Ampoule is a vaccine made from tetanus toxoid, a substance prepared by detoxifying the bacteria Clostridium tetani using formalin and heat. The toxoid is purified and combined with aluminium phosphate. Thiomersal is added as a preservative. The vaccine appears as a greyish-white suspension and does not contain horse serum protein, so Vaxitet Ampoule does not cause sensitivity to equine serum. Vaxitet Ampoule meets the World Health Organization (WHO) standards.
Dosage & Administration
Basic Immunization: Two doses of 0.5 ml, given at least four weeks apart, followed by a third dose 6-12 months later. To maintain immunity, booster doses of 0.5 ml are recommended every 5 to 10 years for adults.
Prevention of Neonatal Tetanus: This vaccine is given to women of childbearing age, especially pregnant women. The vaccination schedule includes three doses initially, followed by two booster doses at least one year apart or during the next pregnancy.
For Injured Persons: If someone has completed their immunization or received a booster in the last 5 years, no extra dose is needed. If more than 5 years have passed and tetanus is suspected, they should receive 0.5 ml of the vaccine immediately. If their immunization history is unclear, they should also receive 1500 IU of tetanus antiserum.
Injection Method: The vaccine should be injected into the deltoid muscle of the upper arm. Only sterile needles and syringes should be used. The vaccine should be shaken well before use. Once opened, multi-dose vials should be kept between 2°C to 8°C.
Reactions
Reactions are usually mild and limited to the injection site. Some swelling, redness, and pain may occur, along with mild fever, tiredness, and irritability. Occasionally, a small bump may form at the injection site. People who have had many booster shots may experience more intense reactions.
Interaction
If passive immunization for tetanus is needed, human tetanus immune globulin (TIG) is preferred. Vaxitet Ampoule provides longer-lasting protection and causes fewer side effects than animal-derived antitoxins. The vaccine should be used cautiously in patients on blood-thinning medications or those with weakened immune systems.
Contraindications
Vaxitet Ampoule should not be given to people who had a severe reaction to a previous dose of tetanus toxoid. Vaccination should also be delayed if the person has a fever or acute infection, though mild illnesses like a cold are not a reason to delay the vaccine.
Side Effects
Most people tolerate the vaccine well. Some common side effects include pain, redness, or swelling at the injection site. Less commonly, people may experience fever or tiredness.
Pregnancy & Lactation
Vaxitet Ampoule is safe to give this vaccine to pregnant women to prevent neonatal tetanus. Vaxitet Ampoule may also be given during pregnancy as early as possible. Vaxitet Ampoule is not known if the vaccine is passed through breast milk, but Vaxitet Ampoule can be given to nursing mothers if needed.
Precautions & Warnings
If anaphylaxis (a severe allergic reaction) occurs after the vaccine, the first treatment is adrenaline. People should be monitored for 30 minutes after receiving the vaccine to check for any immediate allergic reactions. If needed, hydrocortisone and antihistamines should be available.
Reactions may be more severe in people who have received many booster doses. Care should be taken to avoid injecting into a blood vessel.

Storage Conditions
Store the vaccine between 2°C to 8°C, away from light. Do not freeze. If frozen, discard the vaccine. Keep out of reach of children.
