Indications
Xenole 375 is used to relieve the symptoms of osteoarthritis, rheumatoid arthritis, ankylosing spondylitis, and to lower the risk of stomach ulcers in patients at risk of developing NSAID-related gastric ulcers.
Pharmacology
Xenole 375 contains an immediate-release Esomeprazole Magnesium layer and an enteric-coated Naproxen core. Esomeprazole is released first into the stomach, before the Naproxen dissolves in the small intestine.
Naproxen is a type of NSAID that helps reduce pain and fever. Xenole 375 works by stopping the production of prostaglandins. Esomeprazole is a proton pump inhibitor that lowers stomach acid by blocking the final step in acid production in the stomach.
Dosage & Administration
Rheumatoid Arthritis, Osteoarthritis, and Ankylosing Spondylitis: Adults should take one tablet twice a day: either 375 mg of Naproxen/20 mg of Esomeprazole or 500 mg of Naproxen/20 mg of Esomeprazole.
Juvenile Idiopathic Arthritis in Adolescents (12 years and older): For those weighing more than 50 kg: 375 mg of Naproxen/20 mg of Esomeprazole or 500 mg of Naproxen/20 mg of Esomeprazole. For those weighing 38 kg to less than 50 kg: 375 mg of Naproxen/20 mg of Esomeprazole twice a day.
Note: The lowest effective dose for the shortest duration should be used.
Interaction
Xenole 375 may reduce the effectiveness of blood pressure medicines like ACE inhibitors, diuretics, and beta-blockers. Taking Xenole 375 with warfarin can increase the risk of bleeding. Xenole 375 can also interfere with the absorption of some medicines like ketoconazole, iron salts, and digoxin due to changes in stomach acid.
If taken with high-fat food, Xenole 375 can delay the time Xenole 375 takes for Naproxen to be absorbed but doesn’t affect the overall absorption much.
Contraindications
Xenole 375 should not be used by people who are allergic to any of its ingredients, have had allergic reactions to aspirin or other NSAIDs, or during coronary artery bypass graft surgery (CABG).
Side Effects
While Xenole 375 helps reduce stomach ulcers caused by Naproxen, Xenole 375 can still cause some side effects like nausea, vomiting, stomach pain, and indigestion. Long-term use of NSAIDs can be linked to increased risk of heart problems and stomach issues like ulcers or bleeding, especially in older adults.
Pregnancy & Lactation
If pregnant or planning to become pregnant, or if breastfeeding, contact your doctor before using this medicine. Xenole 375 should not be used in the third trimester of pregnancy and should only be considered if the potential benefits outweigh the risks.
Precautions & Warnings
Xenole 375 should be used carefully in people who have a history of stomach ulcers, heart problems, or kidney issues. Older people may experience more side effects, especially stomach bleeding. Xenole 375 should be taken in the lowest dose for the shortest time possible to reduce risks.
Overdose Effects
Overdosing on Naproxen can cause symptoms like stomach pain, dizziness, nausea, and even more serious problems like bleeding or kidney failure. There is no specific antidote for Naproxen or Esomeprazole overdose, and treatment focuses on managing symptoms.
