Indications
Zactrol is used to treat conditions such as gastric ulcer, duodenal ulcer, anastomotic ulcer, acute stress ulcer, reflux esophagitis, and Zollinger-Ellison syndrome. Zactrol is also helpful in treating acute gastritis and chronic gastritis during its worsening stage.
Pharmacology
Zactrol is a histamine H2-receptor antagonist. Zactrol works by blocking histamine’s action on H2-receptors in the stomach, reducing the production of gastric acid. The effect of the drug is reversible over time, as Zactrol slowly detaches from the receptors.
Dosage & Administration
Tablet:
For conditions like gastric ulcer, duodenal ulcer, anastomotic ulcer, upper gastrointestinal bleeding, reflux esophagitis, and Zollinger-Ellison syndrome: The usual dose for adults is 20 mg twice a day (after breakfast and supper or before bedtime), or 40 mg once daily at bedtime.
For treating acute gastritis or chronic gastritis in its acute stage: The usual dose is 20 mg twice a day, or 40 mg once a day at bedtime. The dosage may be adjusted based on age and symptoms. Most ulcer patients heal within 4-8 weeks. For maintenance, the recommended dose is 20 mg once daily or as prescribed by a doctor.

Gastroesophageal Reflux Disease (GERD):
For children:
Under 1 year: 0.5 mg/kg/dose once daily up to 8 weeks.
3 to 11 months: 0.5 mg/kg/dose twice daily up to 8 weeks.
1 to 2 months: 0.5 mg/kg/dose once daily up to 8 weeks.
Neonates: 0.5 mg/kg/dose once daily up to 8 weeks.
For patients 1-16 years of age:
Gastroesophageal Reflux Disease (GERD): 1 mg/kg/day, divided twice a day, up to 40 mg twice a day.
Duodenal ulcer: 0.5 mg/kg/day, divided at bedtime or twice a day, up to 40 mg/day.
Peptic ulcer: 0.5 mg/kg/day, divided at bedtime or twice a day, up to 40 mg/day.
Maintenance therapy: 40 mg daily at night.
Reflux esophagitis: 2 mg/kg/day.
Zollinger-Ellison Syndrome: 40 mg three times a day.
Interaction
No important drug interactions have been found. Zactrol does not affect the cytochrome P450 enzyme system that metabolizes drugs.
Contraindications
Zactrol is contraindicated in individuals who are allergic to any of its ingredients.
Side Effects
Some side effects that may occur are rash, constipation, diarrhea, dry mouth, nausea, vomiting, increased heart rate, high blood pressure, headache, drowsiness, or trouble sleeping. These effects are rare.
Pregnancy & Lactation
Zactrol is classified as pregnancy category B, meaning there are no well-controlled studies in pregnant women. Zactrol should only be used during pregnancy if absolutely necessary. Caution should be taken when giving Zactrol to breastfeeding mothers.
Precautions & Warnings
Zactrol should be used in the lowest effective dose based on the severity of the condition. Special care should be taken when administering Zactrol to elderly patients or those with kidney or liver problems.
