Indications
Zavicefta is used to treat the following infections:
- Complicated intra-abdominal infections (cIAI)
- Complicated urinary tract infections (cUTI), including kidney infections (pyelonephritis)
- Hospital-acquired bacterial pneumonia (HABP) and pneumonia from ventilators (VABP)
Pharmacology
Zavicefta contains two medicines:
- Ceftazidime: This medicine fights bacteria by breaking down their cell walls, which leads to the bacteria dying.
- Avibactam: This helps Ceftazidime work better by stopping certain bacteria from breaking Zavicefta down. Zavicefta works on many types of harmful bacteria, but not all of them.
Dosage & Administration
For Adults with Normal Kidney Function (Creatinine Clearance > 50 mL/min):
- Complicated intra-abdominal infections: 2.5 grams every 8 hours, infused over 2 hours, for 5-14 days
- Complicated urinary tract infections: 2.5 grams every 8 hours, infused over 2 hours, for 7-14 days
- Hospital-acquired bacterial pneumonia: 2.5 grams every 8 hours, infused over 2 hours, for 7-14 days
For Adults with Kidney Problems (Creatinine Clearance < 50 mL/min):
- 31 to 50 mL/min: 1.25 grams every 8 hours
- 16 to 30 mL/min: 0.94 grams every 12 hours
- 6 to 15 mL/min: 0.94 grams every 24 hours
- Less than or equal to 5 mL/min: 0.94 grams every 48 hours
For Children (3 months to less than 18 years):
- 2 years to less than 18 years: 62.5 mg/kg (max 2.5 grams) every 8 hours for 5-14 days
- 6 months to less than 2 years: 62.5 mg/kg every 8 hours
- 3 months to less than 6 months: 50 mg/kg every 8 hours
For Children with Kidney Issues:
- 31 to 50 mL/min: 31.25 mg/kg every 8 hours
- 16 to 30 mL/min: 23.75 mg/kg every 12 hours
- 6 to 15 mL/min: 23.75 mg/kg every 24 hours
- Less than or equal to 5 mL/min: 23.75 mg/kg every 48 hours
Interaction
Zavicefta is not recommended to take this medicine with probenecid (a drug for gout), as Zavicefta may interfere with its effectiveness.
Contraindications
Zavicefta should not be used by people who are allergic to Ceftazidime, Avibactam, or any other cephalosporin antibiotics.
Side Effects
The most common side effects include:
- In adults: Diarrhea, nausea, and vomiting
- In children: Vomiting, diarrhea, skin rash, and swelling or pain at the injection site
Pregnancy & Lactation
There are no well-controlled studies for pregnant women, so Zavicefta should only be used if necessary. The effect on breast milk or breastfeeding is unknown.
Precautions & Warnings
Patients with kidney problems need to be carefully monitored, and the dose may need to be adjusted. Serious allergic reactions, including anaphylaxis, may occur. If any allergic reaction happens, stop using the medicine.
Clostridium difficile infection (CDAD) may occur, causing severe diarrhea. If this happens, consult your doctor immediately.
Patients with kidney problems may experience seizures or other nervous system issues. The dose may need to be adjusted in such cases.
